Testing of Direct Reaction Mixture for Shorter DMTA Cycles through D2B Drug Discovery

Direct-to-Biology (D2B) represents a transformative advance in drug discovery. By testing unpurified reaction mixtures directly in biological assays, D2B drug discovery accelerates the Design-Make-Test-Analyze (DMTA) cycle, reducing time, resource investments, and overall costs.

In this poster, we present Pharmaron’s D2B drug discovery platform for the discovery and optimization of Bifunctional Targeted Protein Degraders (TPDs).

Key Insights from our Direct-to-Biology Poster

  • Established D2B Drug Discovery Platform: We have successfully validated a D2B platform specifically designed for bifunctional targeted protein degraders, supported by a diverse toolbox of E3 ligase binders and linkers.
  • Validated Automation: The  automated direct-to-biology chemistry workflow is validated in both 96-well and 384-well formats, achieving high conversion rates for reaction types like SNAr and amide coupling.
  • High Data Agreement: Our results show excellent agreement between data from unpurified, micropurified, and purified compounds, confirming the reliability and robustness of our D2B drug discovery service.
  • Study Examples: We identify hits and reveal Structure-Activity Relationships (SAR) for BRD4 and AR-FL, demonstrating the practical impact of our platform.
